Main research interests
The Group for the research of tritium labeled amino acids, peptides and proteins, headed by Professor Yurii A. Zolotarev, has carried out a straightforward complex study of solid state catalytic hydration and isotope exchange reactions with participation of spillover tritium, which has formed a new scientific trend in the chemistry of isotope-modified compounds. Yurii A. Zolotarev discovered and investigated the mechanism of the reaction of high-temperature solid-state catalytic isotope exchange (HSCIE) of hydrogen in organic compounds applied on a nonorganic carrier has been studied for the first time. For HSCIE discovered the phenomenon of isotopic equilibrium of gaseous tritium and hydrogen atoms in the molecule of solid organic compound, allowing evenly tritium labeled organic compounds to be produced for the first time, these compounds being previously unavailable. HSCIE is used for synthesis of evenly tritium-labeled peptides for use both in receptor binding analysis and for pharmacokinetics studies. The HSCIE reaction is used in the production of highly tritium-labeled ligands of the brain receptors with parameters higher than commercially available ones. The list of his publications bears over 190 titles, including 24 patents of Russia, USA, France and Great Britain.
1978-1983 г. Synthesized a number of unique polystyrene and polyacrylamide chiral sorbents, allowing the quantitative resolution of racemic tritium labeled amino acids into L and D optic isomers to be performed for the first time (Zolotarev et al. J. Chromatography 1981, 207, 231-236; J. Chromatography 1983, 264 377-384)
1985-2015 г. Discovered for the first time and developed a theoretical description of the HSCIE reaction with spillover hydrogen in amino acids, peptides, proteins and selective ligands of receptors. Highly tritium-labeled compounds with retained physiological activity are obtained by the isotope exchange with gaseous tritium using the HSCIE reaction occurring in solid organic compounds under the action of spillover tritium. A virtually complete absence of racemization makes this reaction a valuable preparative method. (Zolotarev et al. Chemical Reviews, 2010, 110, 5425–5446; Zolotarev et al. Amino Acids 2003, 24, (4), 325-333; Zolotarev et al. J. Phys. Chemistry A, 1999, 103, (25), 4861-4864.; Zolotarev et al. J. Phys. Chemistry C, 2013, 117 (33): 16878–16884)
2002-2006 г. Discovered for the first time and developed a theoretical description of isotopic effects in electron spectroscopy of organic compounds. (Zolotarev et al. J. Molecular Structure (Theochem) 2005 724 (1) 53-59; Zolotarev et al. Amino Acids 2006 31 (4): 403-407)
2005-2015 г. Study of molecular mechanism action of physiologically active peptides. Tritium-labeled peptides are obtained by HSCIE reaction with spillover tritium. The distribution of the isotope label among all amino acid residues of these peptides allows the simultaneous determination of practically all possible products of their enzymatic hydrolysis. (Zolotarev et al. Amino Acids 2006 30 (4): 403-408; Zolotarev et al. Russian Journal of Bioorganic Chemistry, 2006, 32, (2) 166–173; Zolotarev et al. Russian Journal of Bioorganic Chemistry, 2015, 41, (6) 644-656)
